I'm looking at a clean Ubuntu 9.10 install, not Debian.
But, for what it is worth. I inserted
deb http://www.debian-multimedia.org testing main
at the end of /etc/apt/sources.list just as you put it, and it does
work for me, except I did not download their security key

I think this means that the two things are not inherently incompatible
and that there is something funky in the way you are changing the
files so as to break the grammar.
