This is a Mac question.
It belongs in the R-SIG-Mac mailing list (  R-SIG-Mac at r-project.org  )(https://stat.ethz.ch/mailman/listinfo/r-sig-mac)
And it's more an Automator than an R subject.

But to answer your question: just

/usr/bin/Rscript "./test_file/test.R"

will do.
Since there are no spaces in the filepath you can leave out those double quotes.
