[R] a function more appropriate than 'sapply'?

With grep:

wells1 <- wells[grep("^[^\\_]*_[^\\_]*$", wells[,1]),]
wells2 <- wells[grep("^[^\\_]*_[^\\_]*_[^\\_]*$", wells[,1]),]
